Metabolic health

Why it gets attention

  • Weight and body fat
  • Hunger and eating habits
  • Blood sugar
  • Liver fat and fat around the organs
  • Three hormone signals, including glucagon

Retatrutide is called a triple agonist because it acts on three signals in the body: GLP-1, GIP, and glucagon. Researchers are studying how that combination affects hunger, weight, and blood sugar.

What the studies found

Weight loss

In an 80-week study of adults with obesity, the highest-dose group lost 25% of their starting weight on average. The placebo group lost about 4%. TRIUMPH-1 obesity trial

Hunger

In a study of adults with type 2 diabetes, people in higher-dose groups reported less hunger and less tendency to overeat than people taking a placebo. Phase 2 appetite study

Liver fat and deep belly fat

In a smaller study of adults with fatty liver, scans showed a drop in liver fat and in fat around the abdominal organs compared with placebo. Phase 2a liver fat study

Why the third signal matters

GLP-1 and GIP are involved in appetite and blood sugar. Retatrutide also activates the glucagon receptor. Researchers are interested in whether that extra signal changes how the body burns energy and uses fat. Weight loss alone can't tell us how much that pathway contributed. More about the evidence
Why people are interested
Interest centers on appetite, weight, and metabolic health. Retatrutide engages three receptors. GLP-1 and GIP are involved in nutrient and insulin signaling, while glucagon adds a pathway involved in energy and fuel regulation. Human trials test the net effect of engaging all three.
What researchers studied
GLP-1, GIP and glucagon receptor signaling in adults with obesity or overweight and a related condition.
What they found
In TRIUMPH-1, mean weight change at week 80 was −25.0% in the highest-dose arm and −3.9% with placebo. Gastrointestinal events were most common.
Published study methods
TRIUMPH-1 randomized 2,339 adults with obesity without diabetes to weekly study treatment or placebo for 80 weeks. TRIUMPH-2 separately enrolled adults with obesity and type 2 diabetes.
How to interpret the findings
TRIUMPH-1 also assessed knee pain and sleep apnea in prespecified subgroups and reported improvements on those measures. Those outcomes come from participants who had those conditions. The 80-week weight result is a group average from the trial.
TirzepatideTirz · active ingredient in MounjaroHuman randomized trial

Metabolic health

Why it gets attention

  • Weight and body fat
  • Hunger, cravings, and fullness
  • Blood sugar and insulin sensitivity
  • Liver fat and fat around the organs
  • Two hormone signals, GLP-1 and GIP

Tirzepatide is called a dual agonist because it activates two signals in the body. GLP-1 helps regulate appetite, digestion, and blood sugar. GIP also helps regulate insulin. A simple way to remember the difference is that tirzepatide targets GLP-1 and GIP, while retatrutide targets those two plus glucagon.

What the studies found

Weight and body fat

Over 72 weeks, adults without diabetes lost about 15% to 21% of their starting weight on average, depending on the tirzepatide group, compared with about 3% with placebo. In a smaller group who had body scans, both body fat and lean mass fell. SURMOUNT-1 trial · Body composition study

Hunger, cravings, and fullness

In a six-week study, people taking tirzepatide ate less at a test meal and reported less hunger and fewer cravings than people taking placebo. Researchers also asked about feelings of fullness. Appetite and food intake trial

Blood sugar and insulin

Tirzepatide lowered A1C, a measure of average blood sugar over several months. A separate study led by researchers in Germany found that people with type 2 diabetes responded better to insulin, which helps move sugar out of the blood. SURPASS-2 blood sugar trial · German-led insulin sensitivity trial

Deep belly fat and liver fat

In an MRI study of adults with type 2 diabetes, tirzepatide reduced liver fat and fat around the abdominal organs compared with long-acting insulin. SURPASS-3 MRI study

How it compared with semaglutide

In one 72-week trial in adults with obesity without diabetes, average weight loss was 20.2% with tirzepatide and 13.7% with semaglutide. The study compared the two treatments as a whole, so it cannot tell us how much of the difference came from GIP alone. SURMOUNT-5 head-to-head trial

Metabolic health

Why it gets attention

  • How the body breaks down and stores fat
  • Body-fat research
  • Growth hormone fragment and IGF-1
  • Early cartilage research in animals

AOD-9604 is a lab-made version of a small piece of human growth hormone. Researchers designed it to study how the body breaks down and stores fat, separately from the broader effects of growth hormone. Its research started with fat metabolism rather than the GLP-1 appetite pathway.

What the studies found

Why researchers looked at fat breakdown

In an Australian study, rats given AOD-9604 gained less weight, and tests showed that their fat tissue broke down more fat. That result helps explain the interest in AOD-9604, but it came from rats. Australian rat study

An early Australian human trial showed a small weight-loss signal

The developer reported that adults in the 1 mg oral group lost about 2.8 kg (6.2 lb) on average over 12 weeks, compared with 0.8 kg (1.8 lb) with placebo. Higher doses did not consistently do better. These weight-loss results came from the developer’s announcement. 2004 trial results reported by the developer

The larger follow-up did not confirm it

In the next trial, 536 adults enrolled and 502 were randomly assigned to take AOD-9604 tablets or placebo. The groups did not differ clearly in weight loss at 12 or 24 weeks. The developer ended its obesity program. Australian trial registration · Developer’s 2007 results

What the human trials tested

A report covering six Australian trials, with about 900 adults altogether, describes two early studies that gave AOD-9604 into a vein and four that gave it by mouth. The report focused on tolerability and found no rise in blood IGF-1, a growth-related signal, under the conditions studied. Published summary of the six human trials

Later human pain studies under the name LAT8881

The same peptide was later studied under a different name. A UK trial tested oral LAT8881 for nerve pain and found no clear advantage over placebo on its main pain measure. Separate studies tested it for migraine and gave it by IV for pain radiating from the lower back. These studies asked about pain, not weight loss or cartilage repair. UK nerve-pain trial results · Migraine protocol and compound history

IV study for pain radiating from the lower back

An early proof-of-concept trial studied the IV route in volunteers and in adults with pain radiating from the lower back. This trial did not study daily injections under the skin. Registered IV pain trial

Cartilage research

In a South Korean rabbit study, researchers injected AOD-9604 into injured knee joints. Some measures of cartilage damage improved. The experiment tested treatment inside the joint in rabbits. Rabbit knee cartilage study

More about the evidence
Why people are interested
The idea was to isolate a small part of growth hormone and see whether it could influence fat metabolism without reproducing the effects of full-length growth hormone. That distinction is why the compound is discussed in weight and body-composition research.
What researchers studied
Weight change and fat metabolism.
What they found
A study with 536 participants with obesity found no significant weight loss compared with placebo.
Published study methods
Placebo-controlled human obesity research. The larger trial is summarized in the cited regulatory review.
How to interpret the findings
The published analysis of six trials focused on tolerability and found no detectable effect on serum IGF-1. That biochemical finding answers a different question from weight loss. The larger obesity trial did not establish a weight-loss benefit.
5-Amino-1MQ5A1MQ · small molecule · 50 mg vial formatAnimal research

Metabolic health

Why it gets attention

  • Fat-cell metabolism and body composition
  • NNMT and the connection to NAD+
  • Blood sugar and liver fat in animal studies
  • Interest in combining it with GLP-1 treatments

5-Amino-1MQ is a small molecule sold alongside peptides. It blocks an enzyme called NNMT. Researchers are studying whether blocking that enzyme changes how fat cells handle energy and store fat.

What the studies found

What researchers found in people about NNMT

A German study found that fat tissue from adults with type 2 diabetes had roughly twice as much NNMT gene activity as tissue from the comparison group. Higher activity was linked with a weaker response to insulin. Participants were not given 5-Amino-1MQ. German human tissue study

Fat tissue and food intake

In an early study, mice with diet-induced obesity lost weight and had less fat in the tissue the researchers measured. The mice did not eat less overall. This helps explain why the compound is studied separately from appetite, but it does not establish an appetite or weight effect in people. Early mouse study

Body fat, blood sugar, and liver fat

In a later experiment, treated mice gained less body fat at the higher dose and showed changes in blood sugar control and liver fat. This experiment measured outcomes in mice, not people. Follow-up mouse study

The NAD+ connection

NNMT uses a form of vitamin B3 that cells can also use to make NAD+, a molecule involved in turning food into usable energy. Blocking NNMT raised NAD+ in lab-grown mouse fat cells in one experiment. Whether this raises energy expenditure in people remains unknown. Fat-cell experiments

Why people ask about combining it with retatrutide or tirzepatide

Retatrutide and tirzepatide act on receptors involved in appetite and blood sugar. 5-Amino-1MQ blocks an enzyme inside cells. Those different targets explain the interest in pairing them. A federally funded project plans to test a candidate called RT-002 alongside semaglutide in mice. Its public description does not establish that RT-002 is 5-Amino-1MQ. Human results for 5-Amino-1MQ paired with either retatrutide or tirzepatide have not been identified. 5-Amino-1MQ mouse research · Preclinical RT-002 combination research grant

MOTS-cMitochondrial-derived peptideAnimal research

Metabolic health

Why it gets attention

  • Exercise response and physical performance
  • How cells use energy and respond to stress
  • Blood sugar and insulin sensitivity
  • Muscle function with aging

MOTS-c is a small peptide your mitochondria make. Researchers study it as a signal that helps cells respond when energy is low. Its nickname in peptide circles comes from research on some of the same pathways that exercise activates.

What the studies found

Why people call it “exercise in a vial”

In a small human study, naturally made MOTS-c rose in muscle and blood after exercise. In the same paper, mice given MOTS-c performed better on physical tests. The human study measured the body’s own response to exercise. It did not test whether taking MOTS-c improves endurance. Human exercise and mouse performance study

Blood sugar and energy use

Researchers found that MOTS-c can affect AMPK, a pathway that helps cells respond when energy is low. In mice on a high-fat diet, it improved the response to insulin and limited weight gain. This helps explain the interest in how the body handles sugar and fuel. Metabolism study in cells and mice

What human genetics adds

A Japanese-led study found that a naturally occurring MOTS-c variant was linked with higher odds of type 2 diabetes in men, especially those who were less active. The study looked at inherited differences and did not give people MOTS-c. Human genetics study

Why researchers study it in aging muscle

A New Zealand-led study found that older men had less MOTS-c in their blood but more in their muscle than younger men. In a separate experiment, older mice given MOTS-c performed better on physical tests. These findings support further research on aging muscle. Human aging study · Mouse performance study

An early human trial of a related compound

A small trial tested CB4211, a modified compound based on MOTS-c. The developer reported improvements in some blood markers over four weeks. Those early results apply to CB4211 and do not establish a benefit from MOTS-c itself. CB4211 trial registration · Developer’s preliminary results

More about the evidence
Why people are interested
MOTS-c is of interest in exercise and metabolic research because mitochondria do more than make energy. They also send signals that influence how cells respond to stress. MOTS-c is one of the peptides studied in that signaling system.
What researchers studied
Mitochondrial signaling, insulin sensitivity and metabolic stress.
What they found
MOTS-c prevented diet-induced obesity and insulin resistance in mouse experiments. The findings apply to those models.
Published study methods
Controlled rodent experiments used diet and aging models and measured metabolic outcomes.
How to interpret the findings
The original mouse work examined insulin sensitivity and responses to a high-fat diet. Researchers have also measured naturally occurring MOTS-c around exercise in people. A change in an endogenous signal during exercise answers a different question from whether giving the peptide improves performance.

Hormone signaling

Why it gets attention

  • The idea of a brief growth hormone pulse
  • Downstream IGF-1 signaling
  • Interest in lean tissue and fat metabolism
  • Questions about sleep and recovery

CJC-1295 No DAC is commonly used as a name for modified GRF 1–29. It is modeled on the signal that tells your body to release growth hormone. The idea is to encourage a brief signal that fits the body’s natural rhythm.

What the studies found

Why researchers are interested in growth hormone pulses

Growth hormone naturally comes in bursts. In a small Belgian study, giving growth hormone–releasing hormone during deep sleep produced a stronger growth hormone response than when participants were awake. The researchers measured hormone release. They used GHRH itself and did not test whether No DAC improves sleep. Belgian human study of GHRH and sleep

What the human CJC-1295 trials found

The long-acting DAC version raised growth hormone and IGF-1, a signal the body makes in response to growth hormone. In a separate study of that same long-acting version, growth hormone still came in pulses even though levels between pulses were higher. Both studies used CJC-1295 with DAC. Human GH and IGF-1 trial with DAC · Human GH pulse study with DAC

What “No DAC” means

DAC is the added part that makes long-acting CJC-1295 bind to a blood protein and stay in the body longer. The version commonly called No DAC or modified GRF 1–29 leaves out that part. A shorter signal is the idea behind the community’s pulse-style approach. The exact human duration and effects on sleep, muscle, fat loss, or recovery have not been established for this version. Hormone signaling

Why it gets attention

  • Two pathways that signal growth hormone release
  • Interest in brief hormone pulses
  • Questions about lean tissue, fat metabolism, and recovery
  • Interest in sleep and IGF-1 signaling

The blend pairs two compounds that are of interest because they signal the body to release growth hormone in different ways.

10 mg total catalog vial

5 mg CJC-1295 No DAC + 5 mg Ipamorelin.

What the studies found

What ipamorelin did in people

In a study of healthy male volunteers, IV ipamorelin given on its own produced a temporary rise in growth hormone. The study did not measure whether the blend improves muscle, sleep, or recovery. Human study of ipamorelin and growth hormone

Why the pairing is of interest

In a small human study, giving GHRH with a different peptide called GHRP-2 produced a larger overall growth hormone response than either alone. The increase did not clearly exceed the sum of the two individual responses. That study helps explain the two-pathway idea, but used different compounds from this blend. Human study of GHRH plus GHRP-2

Why No DAC matters

The long-acting version of CJC-1295 uses a component called DAC to stay in the body longer. The interest in leaving out DAC is a shorter signal. Human studies have not established the pulse size or duration for this exact blend, or its effects on IGF-1, body composition, sleep, or recovery. Human study of long-acting CJC-1295 with DAC

Hormone signaling

Why it gets attention

  • A brief growth hormone pulse
  • How selectively it affects growth hormone
  • Interest in sleep, recovery, and body composition
  • Why it is paired with a GHRH analog

Ipamorelin activates the ghrelin receptor, a signal that tells the pituitary to release the body’s own growth hormone. Researchers are interested in the brief pulse that follows and how selectively it affects growth hormone.

What the studies found

A growth hormone pulse in people

In a study of healthy male volunteers, IV ipamorelin prompted a single burst of growth hormone. Levels peaked about 40 minutes after administration and then fell. The study measured the hormone response over a short period. Human growth hormone study

Why researchers called it selective

In a Danish animal study, ipamorelin triggered growth hormone release in pigs. Compared with GHRP-2 and GHRP-6, it did not significantly raise cortisol or ACTH, a hormone that tells the body to make cortisol. This comparison was in animals. Danish pharmacology study

Why it is paired with CJC No DAC

Ipamorelin activates the ghrelin receptor. CJC No DAC is designed to act through a different growth hormone pathway. A small human study using other compounds found a larger growth hormone response when both pathways were stimulated. The specific pairing with ipamorelin has not been shown to improve muscle, sleep, or recovery. Human study of related growth hormone signals

More about the evidence
Why people are interested
Ipamorelin is discussed for its ability to stimulate GH release through the ghrelin receptor. Interest in gut motility comes from a separate function of that signaling pathway, which was tested in people after bowel surgery.
What researchers studied
Growth hormone release and postoperative bowel motility.
What they found
Danish experiments characterized selective growth hormone release. A later human trial found no statistically significant improvement in its primary measure of bowel recovery.
Published study methods
The clinical proof-of-concept trial randomized bowel surgery patients to intravenous study treatment or placebo.
How to interpret the findings
The Danish pharmacology work measured hormone responses in laboratory and animal systems. The clinical bowel study evaluated postoperative recovery and did not find a significant benefit on its primary endpoint. Neither design measured muscle gain or sleep quality.

Hormone signaling

Why it gets attention

  • Deep belly fat around the organs
  • Liver fat in adults with HIV
  • Body composition beyond the scale
  • Growth hormone and IGF-1 signaling

Tesamorelin tells the pituitary to release the body’s own growth hormone. The main reason researchers studied it was to see whether it changes where fat is stored, especially the deep belly fat that surrounds the organs.

What the studies found

Deep belly fat

In a 26-week trial of 412 adults with HIV and abdominal fat accumulation, CT scans showed that fat around the organs fell 15.2% with tesamorelin and rose 5.0% with placebo. Triglycerides also fell in the tesamorelin group. Falutz et al. · Visceral-fat trial

Liver fat

In a 12-month trial of people with HIV and fatty liver, tesamorelin lowered liver fat by 4.1 percentage points more than placebo. At one year, 35% of the tesamorelin group and 4% of the placebo group had liver fat below 5%. Stanley et al. · Liver-fat trial

Growth hormone signaling

Tesamorelin mimics the signal that tells the pituitary to release growth hormone. In the 26-week study, IGF-1, a downstream signal, rose in the treatment group. A change in IGF-1 is a lab finding. The trial did not measure workout recovery. Falutz et al. · Human GH pathway and body-fat findings

What the prescription is for

More about the evidence
Why people are interested
Tesamorelin is studied for deep abdominal fat around the organs. As a GHRH analogue it acts through the GH and IGF-1 axis. The pivotal study measured visceral fat by CT, which gives more specific information than total weight.
What researchers studied
Visceral fat in adults with HIV and abdominal fat accumulation.
What they found
At 26 weeks, visceral fat measured by CT fell 15.2% with tesamorelin and rose 5.0% with placebo. This was a defined HIV population.
Published study methods
The double-blind study randomized adults receiving antiretroviral therapy to daily study treatment or placebo for 26 weeks.
How to interpret the findings
Another randomized study in people with HIV and fatty liver measured liver fat over 12 months and found a greater reduction with tesamorelin than with placebo. These studies involved defined HIV populations. They do not establish a benefit for general weight loss or bone density.

Hormone signaling

Why it gets attention

  • The body’s own growth hormone release
  • Nighttime hormone pulses and deep-sleep biology
  • Strength and endurance research in older adults
  • Growth in children with a diagnosed deficiency
  • Interest in body composition, recovery, and IGF-1

Sermorelin mimics the signal that tells the pituitary to release the body’s own growth hormone. Researchers have studied that response in older adults and measured growth in children with a diagnosed growth hormone deficiency.

What the studies found

Growth hormone release at night

In a six-week study of 11 older men, nightly GHRH 1–29 increased overnight growth hormone release and the size of the peaks. IGF-1 did not increase in this study. The hormone measurements tell us about growth hormone release. They do not establish that sleep quality improved. Vittone et al. · Nighttime study in older men

What the adult study found about strength

In that same small study, two of six strength tests and one endurance test improved. These early findings do not establish gains in lean muscle or faster workout recovery. Vittone et al. · Strength and endurance measures

Growth in children with a diagnosed deficiency

In a multicenter study, children with growth hormone deficiency grew faster during their first year of treatment. Among those analyzed, the average growth rate was 4.1 cm per year before treatment and 7.2 cm per year at the 12-month check. That study looked at childhood growth. Why bedtime comes up

The body releases growth hormone in bursts. A human physiology study linked deep sleep with an increase in growth hormone release. This helps explain why bedtime appears in clinical studies and community notes. Van Cauter et al. · Human sleep physiology

Why it gets attention

  • Tendon and ligament healing in animal studies
  • Interest in muscle recovery and wound repair
  • Gut protection and inflammatory signaling
  • Blood-vessel formation and circulation in injury models
  • Early human research on knee pain

BPC-157 is a synthetic peptide modeled on a fragment associated with gastric juice. It is often called a repair peptide because researchers have looked at several parts of healing, including how cells move, how blood vessels form, and how injured tissues respond.

What the studies found

Tendon and ligament healing in rats

Croatian researchers reported improved healing after Achilles tendon or knee ligament injuries in rats. They also found increased growth of tendon cells in the lab. These studies help explain interest in soft-tissue repair. Achilles tendon study · Knee ligament study

An early human report on knee pain

Some patients reported less pain after injections into the knee joint in a small chart review. Some received BPC-157 with thymosin beta-4. There was no comparison group, and the researchers did not measure tendon or ligament repair. Human knee-pain chart review

Gut and intestinal research

An early controlled human study tested BPC-157 as a rectal enema in people with ulcerative colitis. Symptoms and other measures moved in a favorable direction, but the reported results did not establish a clear difference from placebo. The findings were reported in a conference abstract. What they found
Croatian researchers reported improved healing measures in transected rat Achilles tendons and growth of tendon cells in culture.
Published study methods
Surgically injured rats and cell cultures were studied. Human tendon recovery remains unestablished.

Tissue and skin

Why it gets attention

  • Cell movement and tissue remodeling
  • Interest in tendon, ligament, and muscle recovery
  • Wound healing and blood-vessel formation research
  • Heart repair studies of full-length thymosin beta-4
  • Why it is paired with BPC-157 in Wolverine

TB-500 is modeled on a short region of thymosin beta-4, a molecule studied for how cells move and rebuild tissue. The appeal of pairing it with BPC-157 in Wolverine is to explore two parts of repair: tissue protection and how cells move into injured areas. Controlled human trials have not established faster healing with this blend.

What the studies found

Why actin matters

Actin helps cells change shape and move. Researchers studying thymosin beta-4 are interested in whether its actin-binding region helps cells move into injured areas and rebuild tissue. An Italian lab identified a short fragment called Ac-LKKTETQ in a sample sold as TB-500. Identification of the TB-500 fragment

Wound repair in animals

A study in aged mice found improved skin wound repair with a related fragment called LKKTETQ. TB-500 has an added chemical group on that fragment. In a separate lab experiment, TB-500 itself did not improve wound closure under the conditions tested. Mouse wound study · Lab study of TB-500 and its breakdown products

Human wound research with full-length thymosin beta-4

A trial in Italy and Poland studied a gel containing the full-length molecule in 73 people with venous leg ulcers. The researchers reported a possible healing benefit at one concentration. They did not test injectable TB-500. European venous ulcer trial

Why it comes up in heart research

Full-length thymosin beta-4 was studied in heart cells and in mice after heart injury. Researchers observed changes in cell movement and survival. This helps explain the interest in heart repair, but it does not establish that TB-500 improves heart healing in people. Thymosin beta-4 heart repair study

GHK is a short peptide found naturally in the body. When it binds copper, it is called GHK-Cu. Researchers study how that complex affects collagen and the material that gives skin and other tissues their structure.

What the studies found

Collagen and tissue remodeling

Researchers in France found that GHK-Cu helped lab-grown cells make more collagen. In a separate rat wound study, it increased the buildup of collagen and other material that gives tissue its structure. French collagen study · French experimental wound study

Topical wound repair in people

In a controlled study of diabetic foot ulcers, a GHK-Cu gel used alongside standard wound care led to greater wound closure than the gel base alone. The study tested a specific topical product and type of wound. Controlled diabetic ulcer study

What a cosmetic study found

In a small study after laser resurfacing, people using a GHK-Cu skin-care regimen rated their skin more favorably. Independent evaluators found no clear difference in wrinkles or overall skin quality between the groups. Randomized post-laser skin study

The hair research connection

More about the evidence
Why researchers might study this blend
People look into KLOW for skin and connective-tissue repair and for its inflammation angle. The idea is to study different parts of a repair response together: inflammatory signaling, collagen and matrix formation, repair of injured tissue, and movement of cells into a wound. KPV adds the inflammation-focused line of research to the three ingredients in GLOW.
What researchers studied
Inflammatory signaling and tissue-remodeling pathways of individual ingredients.
What they found
The cited work includes mouse intestinal inflammation, fibroblast collagen and rat tendon models. A benefit for this exact blend remains unestablished.
Published study methods
A controlled protocol for the exact combination was not verified.
What the combination could add
The rationale is that the four ingredients address different processes relevant to repair. The studies used different tissues, species, formulations, and endpoints. A direct comparison of KLOW against its individual ingredients would be needed to show that the blend improves an outcome or that its ingredients act synergistically.

KPV

Inflammatory signaling

This three-amino-acid fragment of alpha-MSH was studied for uptake through the PepT1 transporter. Researchers observed less inflammatory signaling in cells and reduced inflammation in chemically induced mouse colitis. That is the basis for KPV’s inflammation angle in the blend.

Cellular health

Why it gets attention

  • Telomeres and telomerase in lab-grown human cells
  • Melatonin and the body clock
  • Interest in sleep-wake regulation
  • Cellular aging and longevity research

Researchers are interested in the protective tips of chromosomes, called telomeres, and whether Epithalon changes them.

What the studies found

Telomeres and cellular aging

Telomeres are protective tips on chromosomes. A Russian research team reported that Epithalon activated telomerase, an enzyme that helps maintain those tips, and lengthened them in human cells grown in the lab. A 2025 study also reported telomere lengthening in cultured human cells. These studies did not measure aging or lifespan in people. Russian telomerase experiment · 2025 human cell study

Melatonin and the body clock in people

In a small study of women with low levels of a melatonin breakdown product, researchers reported changes in that marker and in the activity of genes involved in the body clock after using an Epitalon spray under the tongue. The study measured biological signals, leaving the effect on sleep quality open. Ivko et al. · Human body-clock study · Trial details and route

Why the names matter

Epithalon, also called Epitalon or AEDG, is a synthetic peptide made of four amino acids. Epithalamin is a mixture extracted from pineal tissue. Studies using the extract answer separate questions about that preparation. Evidence review distinguishing both preparations

Why it gets attention

  • Cellular energy and ATP production
  • Mitochondria and metabolism
  • DNA repair and how cells respond to stress
  • Sirtuins and aging biology
  • What human studies measured after an infusion

NAD+ is a helper molecule that cells use to turn food into usable energy. Enzymes involved in DNA repair and stress responses also use it. That connection is why researchers study NAD+ in metabolism and healthy aging.

What the studies found

What happens after NAD+ enters the bloodstream

In a small Australian pilot, researchers tracked NAD+ and related molecules in blood and urine during a six-hour IV infusion. The study showed how quickly the body handled the infusion. It did not test whether people had more energy, recovered faster, or aged more slowly. Grant et al. · Direct NAD+ infusion study

What changes with age

One Australian study found lower NAD+ levels with age in sampled skin tissue. A newer study found no age-related decline in whole blood among healthy adults. The findings depend in part on what tissue researchers measure. Australian human tissue study · Human whole-blood study

What UK researchers found with a precursor

A University of Birmingham-led trial found that oral nicotinamide riboside (NR) changed NAD-related molecules in the muscles of older men. NR is a building block the body can use to make NAD+. The trial did not test NAD+ injections. UK-led human muscle study of oral NR

Cellular health

Why it gets attention

  • How mitochondria make usable energy
  • Cardiolipin and oxidative stress
  • Muscle function and fatigue
  • Research in aging muscle and inherited mitochondrial disease

Think of mitochondria as the engines inside your cells. SS-31, also called elamipretide, is studied for how it interacts with the inner lining where those engines make usable energy. Researchers have explored it in aging muscle and in specific inherited mitochondrial conditions.

What the studies found

How it targets mitochondria

SS-31 interacts with cardiolipin, a fat in the inner lining of mitochondria. That lining helps organize the machinery that makes ATP, the energy cells can use. Lab and animal studies looked at ATP production and damage from oxidative stress. Cardiolipin and mitochondrial research

Energy production in older adults

In a small trial of 39 adults ages 60–85 selected for lower mitochondrial function, one IV infusion improved one measure of how quickly a hand muscle could make ATP immediately afterward. The study did not show a clear improvement in muscle fatigue or a lasting energy benefit. Randomized study in older adults

Walking and fatigue in mitochondrial disease

In a 24-week trial of people with primary mitochondrial muscle disease, elamipretide did not improve walking distance or fatigue compared with placebo. In Barth syndrome, the initial blinded trial also missed its main walking and fatigue goals. MMPOWER-3 trial · TAZPOWER trial

Longer follow-up in Barth syndrome

Ten people entered the follow-up study and eight reached the 168-week visit. At that visit, participants walked about 96 meters farther in six minutes than at the start of follow-up. Fatigue scores were also lower. Everyone received elamipretide, so there was no placebo group during this part of the study. 168-week TAZPOWER follow-up

Kidney blood flow during a procedure

In a 14-person pilot study, people receiving an IV infusion around the time of a kidney artery stent had less evidence of low oxygen in the kidney shortly after the procedure. Blood flow also improved in the treatment group at three months. This was a small study of a specific hospital procedure. Randomized kidney stent pilot study

Dry macular degeneration

In a 176-person trial, elamipretide did not clearly improve the two main measures of low-light vision and retinal damage compared with placebo. Some additional scans suggested slower damage to a layer of light-sensing cells. Those findings need confirmation. ReCLAIM-2 randomized eye study

A specific use in Barth syndrome

In 2025 the FDA granted accelerated approval to Forzinity, a prescription elamipretide injection for people with Barth syndrome weighing at least 30 kg. The decision was based on a measure of knee-extension strength. The approval applies to that product and condition. FDA approval summary

  • Intramuscular research on liver blood tests and semen quality

Glutathione is made from three amino acids. It helps cells handle oxidative stress and supports enzymes the liver uses to process certain compounds. Its central role in these processes is why it is often called the master antioxidant.

What the studies found

Antioxidant levels and immune markers

In a six-month trial of 54 healthy adults, oral glutathione increased glutathione levels in several blood and cell samples. Researchers also saw changes in an oxidative-stress marker and in the lab activity of certain immune cells. The trial measured these markers rather than protection from illness. In a Japanese pilot study of people with fatty liver, a liver enzyme called ALT fell after oral glutathione. There was no placebo group, so the study could not isolate glutathione’s effect. Honda et al. · Japanese liver study

An Italian trial that included intramuscular injections

Adults receiving cisplatin for advanced gastric cancer had less nerve damage in a trial of glutathione. Researchers gave it by IV before chemotherapy and by intramuscular injection on later days. The study tested the combined regimen in cancer care and could not isolate the effect of the intramuscular portion. Cascinu et al. · Italian randomized chemotherapy trial

Why it gets attention

  • Hair follicle growth in lab research
  • Specialized cells that help regulate hair growth
  • Interest in topical scalp and skin formulations
  • Questions about collagen, firmness, and skin remodeling

AHK-Cu is a short peptide that binds copper. It is related to GHK-Cu, and the two differ in their first amino acid. AHK-Cu gets particular attention because researchers studied how it affects human hair follicles and the cells at their base.

What the studies found

Hair follicles and the cells that guide growth

Researchers at Seoul National University exposed isolated human hair follicles to AHK-Cu. The follicles grew longer in the lab, and specialized cells at the base of hair follicles multiplied. The study did not measure visible hair growth on anyone’s scalp. Pyo et al. · Human hair follicles studied in the lab

Why it gets attention

  • Cosmetic research on fine lines and facial expression lines
  • Interest in smoother-looking skin
  • A different research focus from collagen peptides
  • Why it is paired with GHK-Cu and hyaluronic acid

What the studies found

What the human wrinkle study found

A 24-person Korean study compared a patch containing acetyl octapeptide-3 and other ingredients with a hyaluronic acid patch. Some measurements of wrinkles around the eyes improved with the active patch. The study tested the whole microneedle patch and did not isolate the peptide’s effect or test a forehead serum. Brain and sleep

Why it gets attention

  • Interest in focus, attention, and mental clarity
  • Memory, learning, and how the brain adapts
  • BDNF and neuroplasticity in lab research
  • Brain recovery after ischemic stroke
  • Interest in neurotransmitter signaling

Semax is a synthetic peptide modeled on part of ACTH. Much of the interest centers on how brain cells adapt and communicate, from early studies of brain activity in healthy adults to research on recovery after stroke.

What the studies found

Brain network activity in healthy adults

A small Russian study used MRI scans before and shortly after nasal Semax or placebo. Researchers saw a change in a brain network active at rest. The scans measured brain activity rather than performance on a focus or memory test. Lebedeva et al. · Human brain imaging study

Stroke recovery research

Russian researchers followed 110 people after an ischemic stroke and compared rehabilitation groups with and without Semax. They reported changes in blood BDNF and recovery measures. The findings concern people receiving stroke rehabilitation. Gusev et al. · Stroke rehabilitation study

Why BDNF gets attention

BDNF is a protein that helps brain cells adapt and form connections. A study found that Semax changed BDNF and related signaling in the rat hippocampus, a brain region involved in memory. This helps explain interest in learning and neuroplasticity. Dolotov et al. · Rat brain study

Why Semax and Selank are discussed together

Researchers have studied how Semax and Selank each affect brain activity at rest. The imaging study tested them separately, so it does not show whether combining them improves focus. Human imaging study of Semax and Selank

Brain and sleep

Why it gets attention

  • Anxiety symptoms and the stress response
  • Interest in feeling calmer while staying alert
  • Questions about focus, memory, and learning
  • GABA-related signaling and early research on other brain signals
  • Why Selank and Semax are discussed together

Selank is a synthetic peptide modeled on tuftsin. “Calm the noise → sharpen the signal” captures why people discuss it alongside Semax. The research looks at anxiety symptoms and brain signaling, while the effects of combining the two remain open.

What the studies found

Anxiety and the calm-focus idea

In a Russian study of 62 people with generalized anxiety disorder or neurasthenia, researchers reported similar reductions in anxiety symptoms with Selank and medazepam. They also reported less fatigue with Selank. A separate small comparison with phenazepam reported improvement in anxiety and some cognitive measures. These findings come from people with diagnosed conditions and do not establish how sedating Selank is for everyone. Russian comparison with medazepam · Russian comparison with phenazepam

Brain activity in healthy adults

In a study of 52 healthy participants across three groups, researchers saw short-term changes in how some brain regions worked together at rest. Selank and Semax were tested separately. The scans did not establish better focus or memory, or show what happens when both are taken together. Human brain-imaging study

GABA and other brain signals

A rat study found that Selank changed the activity of genes involved in brain signaling, including genes related to GABA, serotonin, and dopamine. This helps explain interest in the stress response. It does not show that Selank raises levels of these brain chemicals in people. Rat study of genes involved in brain signaling

Brain and sleep

Why it gets attention

  • Sleep quality and sleep-wake regulation
  • Interest in deep and restorative sleep
  • Daytime alertness after sleep
  • Stress-response signaling and cortisol research
  • Interest in recovery and pain signaling

DSIP stands for delta sleep-inducing peptide. Researchers were interested in whether it changes how sleep is organized and how people feel the next day. Its name reflects the original research interest in deep sleep.

What the studies found

Sleep and next-day alertness

An early placebo-controlled study of 14 people with severe chronic insomnia reported better nighttime sleep and daytime alertness after DSIP. This was a small, short study. Schneider-Helmert · Early human insomnia study

What an Amsterdam sleep-lab study found

In 16 people with chronic insomnia, researchers reported that those given IV DSIP fell asleep sooner and spent more of their time in bed asleep. Participants did not report better sleep quality, and the authors judged the overall short-term benefit limited. Bes et al. · Dutch sleep-lab study

Stress hormones in a Swedish study

After a single IV dose, a blood measure related to ACTH fell in 11 healthy men compared with saline. Cortisol followed its usual daily decline in both groups. The study does not establish that DSIP lowers cortisol. Bjartell et al. · Swedish stress-hormone study

Hormone signaling

Why it gets attention

  • Sexual desire and related distress in human trials
  • Erectile response in early studies in men
  • Melanocortin signaling and sexual arousal
  • How its research differs from blood-flow treatments

PT-141, also called bremelanotide, is a peptide that activates melanocortin receptors. Researchers have studied sexual desire in women and erectile response in men. In the US, prescription Vyleesi is approved for acquired, generalized hypoactive sexual desire disorder in certain premenopausal women.

What the studies found

Sexual desire and distress in women

Two 24-week trials in premenopausal women with low sexual desire that caused distress found improvements in desire scores and related distress compared with placebo. The number of satisfying sexual events did not clearly increase. RECONNECT phase 3 trials

Erectile response in men

An early study found stronger measured erectile responses after subcutaneous PT-141 in men who reported an inadequate response to sildenafil. A separate study tested a nasal form. Neither study establishes that the Vyleesi dose treats erectile dysfunction in men. Early subcutaneous study in men · Early nasal study in men

Why it differs from sildenafil

Sildenafil acts on a pathway that helps blood flow during an erection. Bremelanotide activates melanocortin receptors. Some of those receptors are involved in brain pathways that researchers study for sexual response. The prescribing information says its exact mechanism for improving low sexual desire is unknown. Vyleesi prescribing information

The Melanotan II connection

Researchers observed spontaneous erections in an early Melanotan II pigmentation study. That observation helps explain why related melanocortin peptides became interesting for sexual-function research. Early Melanotan II human pilot

Why it gets attention

  • How the immune system coordinates its response
  • T-cell and dendritic-cell signaling
  • How the immune system learns to tolerate the body’s own tissues
  • Antiviral responses in chronic hepatitis B
  • Research alongside chemotherapy in melanoma

TA-1 is a naturally occurring peptide associated with the thymus, an organ that helps train immune cells. Researchers study how TA-1 helps coordinate when immune cells respond and when they hold back.

What the studies found

Antiviral response in chronic hepatitis B

In an older randomized trial in Taiwan, 41% of the 26-week group versus 9% of the untreated group cleared two measured viral markers by 18 months after enrollment. None of the responders cleared hepatitis B surface antigen, so the result does not show a cure. Chien et al. · Human hepatitis B trial

Add-on research in melanoma

In an Italy-led European trial, two groups receiving 3.2 mg of TA-1 alongside chemotherapy recorded more tumor responses than the control group. The study did not establish that people lived longer. Maio et al. · European randomized melanoma trial

More about the evidence
Why people are interested
Dendritic cells help decide what the immune system should respond to and pass that information to T cells. Researchers at the University of Perugia in Italy studied how TA-1 changes those signals, including signals involving regulatory T cells, which help keep immune responses in check.
What researchers studied
Immune regulation, antiviral responses and add-on cancer research.
What they found
An older Taiwan trial found a delayed improvement in certain hepatitis B markers after a 26-week course. An Italy-led melanoma study saw more tumor responses in two combination groups, with no clear survival benefit.
Published study methods
Condition-specific trials in people and separate lab and animal studies on immune signaling.
How to interpret the findings
In autoimmune disease, the immune system mistakenly targets the body’s own tissues. Italy-based studies of immune tolerance and an animal model of treatment-related gut inflammation help explain the interest in TA-1. These studies describe how the pathway might work. Clinical benefit for autoimmune disease remains unestablished.

Hormone signaling

Why it gets attention

  • How the brain signals the body to make reproductive hormones
  • LH, FSH and testosterone responses in men
  • Sexual desire and arousal research in men and women
  • Egg maturation during supervised IVF

Kisspeptin is a natural signal that tells the brain to release GnRH. That prompts the pituitary to release LH and FSH, hormones that help control testosterone production, ovulation and fertility.

What the studies found

Testosterone signaling in men

A Scottish study found that a single IV dose of kisspeptin-10 raised LH quickly. Testosterone rose during a longer IV infusion in four healthy men. The single-dose tests did not show a clear testosterone rise. George et al. · Edinburgh hormone study

Sexual desire and arousal

Two UK placebo-controlled studies used short IV infusions of kisspeptin-54 in adults with low sexual desire. Brain responses to sexual cues changed in both studies. In men, researchers also measured a stronger erectile response to sexual videos. These were short laboratory sessions, so lasting changes in libido remain uncertain. Mills et al. · Study in men · Thurston et al. · Study in women

Egg maturation during IVF

UK researchers used one weight-based injection of kisspeptin-54 under the skin at a specific point during monitored IVF. It helped trigger final egg maturation. A later UK trial studied whether a second dose could improve egg yield in patients at high risk of ovarian overstimulation. Jayasena et al. · IVF study · Abbara et al. · Follow-up randomized trial

More about the evidence
Why people are interested
Kisspeptin starts a chain reaction. The brain releases GnRH, the pituitary releases LH, and LH signals the testes to make testosterone. Testosterone replacement therapy supplies testosterone directly. Kisspeptin is still being studied as a way to activate the body’s own signal.
What researchers studied
Testosterone signaling, sexual arousal and egg maturation.
What they found
UK researchers measured short-term hormone and arousal responses and studied a supervised trigger for egg maturation during IVF.
Published study methods
The studies used different forms, routes and participant groups.
How to interpret the findings
The testosterone rise was short term and came from a small IV study. Whether kisspeptin can offer sustained benefits for men with low testosterone or improve male fertility remains open. Sexual-response and IVF trials addressed separate questions.

Cellular health

Why it gets attention

  • Energy metabolism and fatigue linked to low B12
  • Healthy red blood cells and DNA production
  • Brain and nerve function, including the protective coating around nerves
  • Homocysteine metabolism
  • Injectable replacement when absorption is a concern

Vitamin B12 helps your body make red blood cells and DNA, keep nerves healthy, and use nutrients for energy. Low B12 can contribute to tiredness, weakness and trouble concentrating. Replacing missing B12 can help deficiency-related symptoms. Extra B12 has no proven energy benefit when levels are already adequate.
